size of 150 (p >0.05). The cost of the consumables for the assay is approximately 10% less expensive than the dot blot assay. A plate containing 80 samples can be analyzed in under 30 min by the fluorescence-based assay, an eighth of the time required per sample for the dot blot; a single operator can easily assay over 1100 samples per day, compared to 240 samples by dot blot. Furthermore, the assay is readily automatable using a proprietary robotic workstation. The assay has not yet been fully validated for forensic casework samples and is used solely on samples submitted for intelligence databasing purposes.

Targeted DNA Methylation Analysis by Next-generation Sequencing
The role of epigenetic processes in the control of gene expression has been known for a number of years. DNA methylation at cytosine residues is of particular interest for epigenetic studies as it has been demonstrated to be both a long lasting and a dynamic regulator of gene expression. Efforts to examine epigenetic changes in health and disease have been hindered by the lack of high-throughpu...
متن کاملA Simple Genome Walking Strategy to Isolate Unknown Genomic Regions Using Long Primer and RAPD Primer
Background: Genome walking is a DNA-cloning methodology that is used to isolate unknown genomic regions adjacent to known sequences. However, the existing genome-walking methods have their own limitations. Objectives: Our aim was to provide a simple and efficient genome-walking technology. Material and Methods: In this paper, we dev...
متن کاملShotgun DNA microarrays and stage-specific gene expression in Plasmodium falciparum malaria.
Malaria infects over 200 million individuals and kills 2 million young children every year. Understanding the biology of malarial parasites will be facilitated by DNA microarray technology, which can track global changes in gene expression under different physiological conditions. However, genomes of Plasmodium sp. (and many other important pathogenic organisms) remain to be fully sequenced so,...
متن کاملQuality Control of High-Throughput Library Construction Pipeline for KAPA HTP Library Using an Agilent 2200 TapeStation
Next Generation Sequencing requires the input of high molecular weight genomic DNA to construct quality libraries for whole genome bacterial sequencing. Large scale sequencing projects, such as the 100K Pathogen Genome Project, require methods to rapidly assess the quantity and quality of the input DNA using high-throughput methods that are fast and cost effective. In this study, the Agilent 22...
متن کاملCapturing native long-range contiguity by in situ library construction and optical sequencing.
The relatively short read lengths associated with the most cost-effective DNA sequencing technologies have limited their use in de novo genome assembly, structural variation detection, and haplotype-resolved genome sequencing. Consequently, there is a strong need for methods that capture various scales of contiguity information at a throughput commensurate with the current scale of massively pa...
